内皮素、一氧化氮、烫伤血清对大鼠心脏微血管内皮细胞ICAM-1表达的影响 被引量:1

摘要 目的:观察内皮素(ET)、一氧化氮(NO)合成酶抑制剂L-NAME以及烫伤血清对心脏微血管内皮细胞细胞间粘附因子ICAM-1表达的影响。方法:分离、培养的SD大鼠心脏微血管内皮细胞分别用ET、L-NAME、烫伤血清刺激后ELISA测定内皮细胞ICAM-1的表达。结果:ET、L-NAME、烫伤血清均能明显诱导微血管内皮细胞ICAM-1的表达。ET在刺激后2h达到高峰,是正常对照的4.55倍;L-NAME刺激后的表达高峰在1h,是正常的3倍。烫伤血清刺激后ICAM-1在4h后表达最强,为正常的3.5倍。结论:烫伤血清、ET、NO均参与、介导大鼠心脏微血管内皮细胞ICAM-1的表达,但表达高峰不在同一时相。 Objective:To observe the effect of endothelin(ET),nitric oxide (NO) and burned rat serum on the ICAM-1 expression of rat cardiac microvascular endothelial cells.Methods:The ICAM-1 expression was measured using ELISA after the endothelial cells were incubated with ET,L-NAME and burned rat serum respectively in different time. Results:The ICAM-1 expression increased significantly after stimulation.The most dramatic rise of ICAM-1 expression was at 2h,1h and 4h after incubation with ET.LNAME and burned rat serum respectively, which was 4.55,3, 3. 5 times as much as control.Conclusion:ET,NO might play a role in the ICAM-1 expression of rat cardiac microvascular endothelial cells which might participate in the injury of endothelial cells after burns.
