摘要
目的:探索表皮细胞在诱导供体皮肤特异性延长成活中的作用。方法:C57BL/6小鼠移植BALB/c小鼠皮肤前1d和植皮后2d腹腔内注射兔抗小鼠T淋巴细胞抗血清0.5ml。植皮后7d胸腺内注射BALB/c小鼠皮肤细胞5×106个或(和)脾细胞1.25×107个。结果:供体表皮细胞和脾细胞共同注射到小鼠胸腺内,结合短暂的免疫抑制,可以使BALB/c。小鼠皮肤成活明显延长,11只鼠中有8只异体皮片平均成活时间达68d。而C57BL/6小鼠胸腺内注射BALB/c小鼠的皮肤细胞和脾脏细胞不能显著延长DBA小鼠皮片成活。若单纯经抗血清处理或胸腺内注射脾细胞或皮肤细胞,虽然也可以延长BALB/c小鼠皮片成活,但时间未超过35d.结论:(1)结合短暂的免疫抑制,受体胸腺内注射供体表皮细胞和脾细胞可以诱导供体皮肤特异性延长成活;(2)皮肤中存在着影响皮肤移植耐受诱导的抗原。
Objective:To observe the effect of the donor epidermal cells and splenocytes on the induction of skin specific unresponsiveness in their injection into the thymus of a mouse immunomodulated with rabbit anti-mice T lymphocytes serum(ALS).Methods:C57BL/6 mice received ALS(0.5 ml) on d-1 and +2 relative to BALB/c skin grafts on d 0,then 1.25×107 splenocytes and/or 5×106 epidermal cells by intrathymic(IT)injection on d 7.Results: IT injection of BALB/c epidermal cells and splenocytes significantly prolonged allografts survival,with 8 of the 11 skin grafts average surviving 68d.and simultaneous rejected MHC non-relative third party DBA skin graft. However,none of the BALB/c skin graft survival reached over 35 d by only ALS injection intraperitonealy or IT injection with only epidermal cells or only splenocytes. although they could prolong BALB/c skin graft survival.Conclusion:Skin specific unresponsiveness could be induced by IT injection of allogenic splenocytes and epidermal cells ;and there is specific antigen affecting the induction of skin specific unresponsiveness in skin.
