“瘢痕Ⅱ”对人皮肤瘢痕成纤维细胞增殖的抑制作用

Inhibitory effect of "Scar Ⅱ"on growth of human fibroblasts in skin scar

目的：观察＂瘢痕＂对皮肤瘢痕组织中成纤维细胞增殖的影响，为该药临床应用提供依据。方法：分别取人皮肤瘢痕组织和正常人表皮组织进行原代细胞培养，加入倍比稀释的＂瘢痕Ⅱ＂，定时观察细胞形态，用ＮＡＧ酶法测定细胞增殖活性。结果：＂瘢痕Ⅱ＂浓度＞０．５ｍｇ／ｍｌ时，生长的表皮细胞膜片部分脱落；成纤维细胞对＂瘢痕Ⅱ＂较敏感，加药各组成纤维细胞的形态发生了明显变化，细胞变圆、减少，药物浓度＞０．４ｍｇ／ｍｌ时，成纤维细胞全部失活；培养３ｄ时，药物浓度与表皮细胞及成纤维细胞活性Ｄ值均成负相关。结论：＂瘢痕Ⅱ＂对人皮肤瘢痕成纤维细胞和人正常表皮细胞的生长均有抑制作用，尤其对成纤维细胞生长抑制效应更为明显。

Objective:To explore the effect of the drug 'Scar Ⅱ'on growth of fibroblasts in skin scar and keratinocytes in normal skin and provide basis for clinic application of this drug.Methods:The scar tissue and normal skin were scissored to pieces and cultured in DMEM containing supplements. The fibroblasts were collected after the fibroblasts grew out.Appropriate amount of the drug 'ScarⅡ' was added . the cell shape was observed continuously and the cell growth was measured with NAGase assays. Results : As the concentration of'Scar Ⅱ'≤0.25 mg/ml, there was not any significantly inhibitory effect on keratinocyte growth. As the concentration>0. 5 mg/ml , the keratinocyte sheet partly detached.Fibroblasts were sensitive to 'ScarⅡ'. As the concentration of 'Scar Ⅱ' >0.4 mg/ml,fibroblasts died completely.After cultured for 3 d. there was a negative correlation between concentration of 'Scar Ⅱ'and keratinocyte(or fibroblast) growth. Conclusion: There is significantly inhibitory effect of 'ScarⅡ'on fibroblasts in skin scar and keratinocytes in normal skin , particularly on fibroblasts.
