摘要
本研究旨在探讨川芎嗪对离体大鼠肝细胞 Ca^(2+)摄入的影响。采用胶原酶原位灌流肝脏方法分离获得大鼠肝细胞,用^(45)Ca^(2+)示踪法观察肝细胞对^(45)Ca^(2+)的摄入与释放过程。结果表明,在无药物存在情况下,肝细胞快速摄入^(45)Ca^(2+),16min 时达最大负荷,然后迅速释放钙;应用川芎嗪则细胞内^(45)Ca^(2+)浓度较低。提示川芎嗪具有抑制肝细胞摄入Ca^(2+)的作用。
The purpose of the present study is to evaluate the effect of Ligustrazin on uptake of Ca^(2+)in isolated rat hepato- cytes.Rat liver was perfused in situ via the portal vein with buffer containing collagenase and isolated liver cells were ob- tained.The uptake and release of ^(45)Ca^(2+)by hepatocytes were investigated by testing tracer ^(45)Ca^(2+).The results indicated that:(1)the uptake of ^(45)Ca^(2+)by hepatocytes was rapid and the concentration of ^(45)Ca^(2+)reached its peak 16 min after in- cubation without Ligustrazin,then ^(45)Ca^(2+)were released rapidly.(2)the concentration of ^(45)Ca^(2+)in hepatocytes was lower after using Ligustrazin.It suggests that Ligustrazin has effect on the uptake of Ca^(2+)in isolated rat hepatocytes.
