人胚脑提取液对神经细胞生长影响的研究

The Effect of Fetal Brain Extrac on the Growthof Cerebral Cortex Neurons in Vitro

用水囊引产3～5个月胎龄的人胚大脑皮层神经细胞进行培养,观察不同浓度(蛋白量0.05～0.4g/L)人胚脑提取液(FBE)对大脑皮层神经细胞生长的影响.结果表明,培养14天神经细胞存活数在0.05g/L和0.1g/L FBE组与对照组(培养液中不加FBE)之间无明显差异,0.2g/L FBE组明显高于对照组,而0.4g/L FBE组存活细胞少于对照组.进一步观察可见0.2g/L FBE组神经细胞突起数目、长度、胞体面积均高于对照组(P<0.05),神经细胞存活天数及细胞总蛋白含量明显增加.结果提示,0.2g/L FBE对体外培养大脑皮层神经细胞的生长有一定促进作用.

The effect of fetal brain extract(FBE)on the growth of cerebral neurons from hu-man fetal brain was investigated by using dissociated cellt culture. Concentrations of proteinin FBE in culture medium was 0. 05g/L. 0.1 g/L.0. 2g/L and 0. 4g/L respectively. On the14th day, the number of survival neurons in 0. 2g/L FBE group were signifcantly higher thanthose of control group (P<0. 05), and survival neurons in 0. 4g/L FBE group were lowerthan those of control group. The number and length of neuronal processes as well as the areaof neurosma in 0. 2g/L FBE group were all increased markedly as compared with those ofcontrol group. The survival days of neurons and the total amount of protein of cells were al-so increased markedly. These results indicated that the suitable amoht of FBE supplementa-tion could improve neuronal growth in vitro.
