用ELISA和PCB对猪、牛血清中HBV抗原、抗体及DNA的检测

DEIECTION OF HBV-Ags、-AbS AND -DNA IN SEBA FROM DOMESTIC ANIMALS USING ELISAAND PCR

用三个厂家生产的乙型肝炎（ＨＢ）ＥＬＩＳＡ试剂盒及聚合酶链反应（ＰＣＲ）对牛、猪和羊血清分别进行了乙型肝炎病毒抗原（ＨＢＶ－Ａｇ）、抗体（Ａｎｔｉ－ＨＢＶ－Ａｂ）及ＤＮＡ的检测。ＥＬＩＳＡ共检出阳性牛血清１３份（１３／１３６），其中ＨＢ＿ｓＡｇ阳性８份，ＨＢ＿ｅＡｇ阳性５份；阳性猪血清４份（４／３６），其中有ＨＢ＿ｓＡｇ阳性３份，ＨＢｅＡｇ阳性１份。全部阳性血清和部分阴性血清共１２０份经用ＰＣＲ检测ＨＢＶＤＮＡ均为阴性，未扩增出特异性ＤＮＡ片段。此结果初步表明，无论家畜血清中有无ＨＢＶ－Ａｇ，均无感染性ＨＢＶ粒子存在，因此没有足够的证据担心家畜会在ＨＢＶ的传播上对人类构成威胁。

Hepatitis B virus (HBV)antigens(HBV-Ags)、antibodies against HBV(HBV-Abs)and DNA in sera from cattle、pig and sheep were determined using ELISA and Poly-merise Chain Reaction(PCR).Of 132 cattle serum samples,8 were HBsAg positive and 5HBeAg positive. 3 of 36 porcine serum samples were HBsAg positive and one of themwas HBeAg positive.None of 8 sheep serum samples was found positive to any HBV-Ags or -Abs in ELISAs.For all cases, whether they were positive or negative in ELISAs, no HBy DNAfragments were amplificated by PCR.This indicates that no infectious HBV exists inthe sera although some of them were positive in ELISAs.Therefor,the evidence toworry about that the livestorck can threaten mankind in the spread of HBV is notsufficient.
