摘要
设计老龄大鼠的急性部分肝胆管梗阻感染(AOPH)模型,并以累计死亡率、腺苷酸及能荷为指标,研究脓毒症和非脓毒症状态下不同月龄(6、12.18和24)大鼠肝细胞的能量代谢。结果显示,随着月龄的增加,肝脏ATP、ADP和总腺苷酸含量逐渐降低,24月龄组分别降为6月龄组的72.7%、68.3%和76.2%。AOPH24h肝细胞能量代谢受到明显破坏,表现为ATP含量减少,AMP含量增加,总腺苷酸减少,能荷水平降低;AOPH48h时更加明显,能荷多低于0.50,大鼠死亡率明显上升。以上改变的突出特点是直接受损的肝左叶较间接受损的肝右叶更加明显,高龄组较低龄组更加明显。提示老化肝脏肝细胞能量代谢低下且容易遭受破坏是其储备功能降低和受损易感性升高的重要机制。
Model of sepsis after acute obstructive partial hepatocholangitis (AOPH) was designed with the parameters including adenine nucleotides and energy charge(EC). Hepatocyte energy metabolism of rats of various mo old(6,12, 18 and 24) in endotoxemic and non-endotoxemic states were investigated. The result showed that the liver ATP. ADP and gross adenosine phosphates(GAP) contents were decreasing with age, with the 24 mo old group were reduced to 72.7%, 68. 3% and 76.2% respectively as compared with the 6 mo old group. The hepatocyte adenosine metabolism was evidently damaged at AOPH 24h and 48h, manifested by reductions of ATP and EC and increase of AMP and characterized by the fscts that the damaged energy metabolism was more evident in the directly artacked left lobe than in the indirectly attacked right lobe which was more easily occurred in elderly rats. The results indicate that the hepatocyte energy metabolism of aged liver was slightly damaged and more easily to suffer. which may be one of the important factors related to the decreased reserve and the increased vulnerability of the aged liver.
