PHA及rhIL-2对新鲜分离肝癌细胞体外实验观察

摘要 本文从本院外科8例肝脏病人,合并肝门静脉高压行肝叶切除的肝组织,分离培养肝癌细胞。其中4例见肝癌细胞生长。对其中1例新鲜分离肝癌细胞中加入植物血凝素(PHA)及重组人白细胞介素-2(rhIL-2),培养3天后发现,原来成片成堆生长的肝癌细胞逐渐减少,细胞结构破坏,7-9天后在细胞消失,周围淋巴细胞增多。未加PFA及rhIL-23例肝癌细胞生长旺盛,而周围淋巴细胞却逐渐减少消失。提示:PHA及,rhIL-2对肝癌细胞周围淋巴细胞有刺激生长作用,对自体肝癌细胞有抑制作用。 ight cases of surgical hepatectomy tissue culture in vitro:5 cases of primary carcinoma of theliver,3 cases of hepatocirrhosis anid one case of liver hydatid disease All cases were complicated byhepatoportril hypertensive,In the 4 cases specimens with liver cancer cells growth,the diagnosesof clinical and pathologic biopsy conformed to primary liver carcinoma. We investgated the effectof phytohemagglutinin(PHA)and recomhinant human interluekin-2(rhIL-2)on the in vitrogrowth of freshly isolated liver cancer cells from one of the speciment with liver cancer cellgrowth,using PHA stimulated previously and then rhIL-2 activated the tumor infiltratinglymphcyte(TIL),it showed that liver cancer cells growth decreased after 3 days of culture andafter 7-9days almostly disappeared,however,TIL increase .Our study deonstrate the combinantionof PHA and rhIL-2 with liver cancer cells growth in vitro has stimulating effect on TIL and aspecific reduction effect on autoliver cancer cells.
