摘要
目的建立一种骨组织工程研究中简便易行的标准化小鼠股骨临界骨缺损模型。方法测量小鼠股骨的解剖参数。将30只2~3月龄FVB/N品系的小鼠分成3组,每组10只,用我们自行设计的内固定物固定后分别制备长度为1、2 mm和2.5 mm的股骨干中段骨和骨膜缺损模型,通过影像学和组织学的方法来观察骨缺损的愈合进程。结果术后3个月通过X线片和组织学检查证实1 mm的缺损可以通过自身修复机制完成骨缺损的愈合,而2、2.5 mm的缺损则无法自行愈合,建模成功率为89.7%。结论通过自行设计的内固定物构建的小鼠股骨缺损模型,在不填充植骨材料的情况下,2 mm以上的骨缺损不能自行愈合,符合临界骨缺损的标准。
Objective To establish a simple,reproductive and standardized method for mouse femur critical bone defect for research in tissue engineering.Methods Anatomic data of mouse femur were obtained through measurement in order to guide the operation.Thirty 2 to 3 months old FVB/N mice were randomly divided into 3 experimental groups,each group with 10 mice.Their right femur were inflicted to make a defect in sizes of for 1,2 and 2.5 mm respectively after the mice were fixed in our self-designed fixation equipment.Bone defect healing process was assessed by plain X-ray filming and histological observation.Results X-ray film and histological observation confirmed that the bone defect at the size of 1 mm was restored by the self-repairing mechanism in 3 months after operation,but no such repair were found in the defect of 2 and 2.5 mm groups.Satisfactory models were created in 89.7% experimental mice.Conclusion Mouse femur critical defect model can be established through our self-designed fixation equipment.Bone defect in size of 2 mm and more in mouse femur can not heal up spontaneously even in 3 months after operation.This defect model has high repeatability and can be used for the related research in tissue engineering as standard animal models.
