MTT法和双琼脂MTT法测肿瘤药敏及其临床应用

CLINICAL USEFULNESS OF CHEMOSENSITIVITY TESTING USING THE MTT METHOD AND DOUBLE-LAYER AGAROSE MTT METHOD

本实验用MTT法测定了258例恶性肿瘤对17种常用抗癌药的体外敏感性,其中30例用MTT法和双琼脂MTT法同时测定,结果显示后者抑制率高于前者.有25例用MTT法测定的实体瘤可评价其近期疗效,其中11例药敏试验显示不敏感而临床化疗无效,另14例抗癌药体外敏感而临床化疗8例有效,抗癌药体外敏感性和临床疗效总符合率为80%.

We tested the in vitro chemosensivity of 258 malignant tumours with about 17 antitumor drugs using MTT assay, and tested 30 patients using both MTT method and dou-ble layer agarose MTT method. It demonstrated that inhibition rate in double-layer agarose was higher. We evaluated the 25 patients with remaining measurable tumor lesions, the 11 patients' antitumor drugs were resistence in vitro, the clinical results were ineffective, the other 14 patients' antitumor drugs were sensitive in vitro and 8 patients' clinical results were effective. The overall prediction rate was 80%.
