摘要
建立了快速、简便测定人血清中沙丁胺醇的反相HPLC荧光法.采用硅胶小柱固相提取,流动相为水相:甲醇=68:32,其中水相为磷酸缓冲液(5.8mmol,pH=6.00)含硫酸庚烷钠5mmol,内标为间羟胺,在0.75~30ng/ml浓度范围内有良好的线性关系,最低检测浓度为0.5ng/ml.在高、中、低浓度时的相对回收率分别为103.0%,97.9%,99.07%.用本法测定了人po沙丁胺醇后的体内血药浓度,经计算机拟合符合二室开放模型,并进行了药动学参数计算.
A fast,simple and accurate method for determining Albuterol Sulfate concentrations in human plasma has been developed and validated for use in routine clinical analyses. This method involves a solid-phase extraction procedure using silica cartridges and reverse-phase high-performance liquid chromatography with fluorescence detection. Its mobile phase was composed of phosphate buffer solution(5.8mM,pH=6.0. containing sodium 1-heptanesulfonate 5mM) : methane = 68: 32. Metaraminol was used as the internal standard. The average relative recoveries of 2. 5,10,and 30ng/ml were 103. 0% ,97. 9% and 99. 07%re-spectively. Regression analysis showed the method was linear in the standard curve range from 0. 75ng/ml to 30ng/ml.The present assay procedure has been applied for monitoring plasma levels in oral phar-macokinetic studies in human body. The best fit was found to be with a two compartment open model. The results imply that Albuterol is absorbed rapidly,distributed widely in body and also eliminated at a fairly rapid rate.
