摘要
应用聚合酶链反应(PCR)方法检测了ANLL,CML,T-ALL,c-ALL和CLL等共119例病人的白血病细胞和14例正常人外周血单个核细胞中T细胞特异转录因子GATA3的表达情况。在T-ALL,c-ALL,CLL及CML中,其表达率很高,分别为75%、85.71%、100%及76.47%;在ANLL中的表达率则为42.68%;在正常外周血中为28.57%。同时,分析了ANLL各亚型GATA 3基因的表达情况,各型间的阳性率没有显著性差异(P=0.3118)。本结果提供了GATA 3基因在白血病中异常表达的情况,提示该转录因子在白血病细胞的分化过程中可能起到一定的调节作用。同时,由于该基因在白血病中的高表达现象,有应用于临床作为检测微小残留病变的价值。
The expression of T cell specific transcription factor GATA 3 in leukemic celis from 119 cases with ANLL, CML, T-ALL, c-ALL, CLL and peripheral blood mononuclear cells (PBMC) from 14 normai subjects was investigated with polymerase chain reaction (PCR) meth-od. The expression rates of GATA 3 in T-ALL, c-ALL, CLL and CML were high: 75%, 85.71%, 100% and 76.41%, respectively, while 42.68% in ANLL and 28.57% in normai PBMC. The expression of GATA 3 in each ANLL subtype was analysed and no statistic signifi-cance was found. Our data indicated that there was aberrant expression of GATA 3 in leukemia and suggested that the transcription factor may play a regulatory role in differentiation process of leukemia celis. High Ievel expression of GATA 3 gene could be applicable to detection of minimal residual disease.
