应用PCR方法检测淋巴细胞白血病IgH和TCRγ基因重排 被引量:1

DETECTION OF IgH AND TCR_γ GENE REARRANGEMENT IN LYMPHOCYTIC LEUKEMIA BY POLYMERASE CHAIN REACTION

摘要 本文应用PCR方法扩增IgH基因重排产生的CDR-Ⅲ区序列和TCRγ基因重排产生的V_9区序列,共检测了20例淋巴细胞白血病病人,其中15例B淋巴细胞白血病均发生IgH基因重排,并有2例出现两种基因扩增产物,它的大小为70-140bp;另5例T淋巴细胞白血病中有2例发生TCRγ基因重排,扩增产物的大小为170-230bp,而在正常人和非淋巴细胞白血病中均未发现有IgH和TCRγ基因重排。 Polymerase chain reaction(PCR) was used to amplify the complementarity determining region 3(CDR-3) of immunoglobulin heavy chain gene(IgH) and the V9 region of T-cell receptor γ chain gene (TCRγ) . Twenty patients with lymphocytic leukemia were detected. IgH rearrangement could be shown in 15 patients with B lymphocytic leukemia, 2 cases of them presented 2 kinds of gene amplification products with the length of 70-140bp. TCRy rearrangement was detected in 5 cases with T lymphocytic leukemia, 2 cases of them presented specific gene amplification products with the length of 170-230bp. None of IgH and TCRy rearrangement were shown in normal and nonlymphocytic leukemia.
