摘要
本文研究了硫酸葡聚糖(DS)的不同含硫量与过敏反应的关系,初步找到了一个可供人体静脉注射用的DS含硫量的范围。DS的分子量在10000以下,这个范围大约为10.6%~11.8%(以进口的葡聚糖为原料)和12.0%~13.0%(以国产葡聚糖为原料)。初步探讨了影响小鼠外周血白细胞变化的某些因素,为DS作为造血干细胞动员剂用于临床展现了良好的前景。
In this paper the relationship 'between the sulphur content in dextran sulphate (DS) and anaphylactic reaction was studied in guinea pigs. We primarily found that the sulphur content in DS which meets the need for the intravenous injection in human was approximately 10.6%-11.8% (for imported dextran) and 12.0%-13.0% (for home-made dextran ) respectively while the molecular weight of DS was less than 10 000. The sulphur content at these levels had also remarkable mobilization effect in mice, especially when DS (home-made) was purified. At 3h after injection the number of pheripheral WBC elevated by 2.41 times more than that in controls injected with NS. Some factors affecting the mobilizing effect in mice and anaphylactic reaction in guinea pigs were investigated. This result provides a fine prospect for DS as a hematopoietic stem cell mobilizer in clinical trials.
