摘要
应用斑点杂交、放免测定等方法检测分析了内毒素不同浓度同时相(A组)和内毒素同浓度不同时相(B组)作用下体外培养的新生牛肺动脉内皮细胞ET—1基因表达和释放的作用,且该作用有明显的剂量依赖关系和时间效应关系,两组ET—1基因,表达与释放。结果表明,内毒素具有促进内皮细胞ET—1基因表达的强弱与释放量的增减均呈显著的正相关。提示内毒素血症时血浆ET水平升高与ET合成增加和基础释放量增高有密切关系。
In this experiment.it was studied the effects of E Coli. endotoxin at different concentrations and times on the gene expression of the endothelin-(ET-1) and its release in bovine pulmonary arterial endothelial cells (PAEC) with the blot hybrid and radioimmunoassay. The results showed that the endotoxin stimulated the gene exression and release of ET - 1 in PAEC in a dose and time dependent way. The gene expression of ET-1 had a positive correlation to the release of ET-1 in the plasma of patients with endotoxemia was induced by the increase of the synthesis with endotoxemia was induced by the increase of the synthesis and relrase of ET-1 in the endothelial cells.
