来复汤对大鼠离体心脏乳头肌肌力效应的影响

Inotropic Effect of Laifu Decoction(来复汤)on Isolated Rat Heart Papillary Muscle

来复汤为近代名医张锡纯用于救治“厥脱证”的常用方剂,为探明其“救脱”的现代药理机制,本实验用离体心脏乳头肌灌流的方法,观察了该方对大鼠心脏肌力效应的影响。结果表明:该方对大鼠离体左室乳头肌有显著的正性肌力效应(P<0.05),β受体阻断剂心得安对这一效应无明显影响,钙通道阻滞剂异搏定能明显拮抗这一效应。提示来复汤对在体心脏亦可能有增强收缩力的作用,且这一作用的机理可能与兴奋心脏的β受体无关而与心肌细胞钙通道的开放及钙离子的转运有关。

Laifu Decoction(LFD)is a recipe for rescuing the urgent cases of collapse by Dr.Zhang Xi-chun,a well- known TCM physician of modern times.In order to find out its anticollapse mechanism in modern pharmacology, a perfusion experiment of isolated rat ventricular papillary muscle was made to investigate the inotropic effect of LFD on the rats'heart.Results showed that LFD had a dramatic postive inotropic effect on the isolated rats'left ventricular papillary muscle(P<0.05).This effect was not influenced by propranolol(a beta-adrenergic blocker),but verapamil(a calcium-channel blocker)obviously antagonized it.This suggested that LFD might also exert a positive inotropic effect of the heart in vivo.The inotropism might be irrelevant to beta-adrenergic receptor which was often connected with the heart stimulation,but was relevant to the calcium-channel opening and calcium-ion transportation in the myocardial cells.
