川芎嗪防治模拟高原犬呼吸窘迫综合征实验研究 被引量:1

Experimental Study of Tetramethylpyrazine on Prevention of High Altitude Simulating Respiratory Distress Syndrome in Dogs

摘要 21只犬在模拟4000m 高原随机分为对照组、致伤组及川芎嗪治疗组,动物均于伤后6h 活杀。结果显示,川芎嗪可抑制犬呼吸窘迫综合征(RDS)伤后外周血 WHC 下降,减轻肺内炎性细胞浸润及肺水肿,但对动脉血氧分压(PaO_2)下降无明显改善。其机制与抑制炎性细胞功能。即抑制多核白细胞(PMN)和肺泡巨噬细咆(AM)之白细胞三烯 B_1合成及 PMN 释放血小板激活因子活性,保护肺血管内皮细胞有关。 Twenty-one hybrid dogs in simulating a high altitude environment of 4000m,were randomly divided into respiratory distress syndrome(RDS)treated and control group.All animals were sacrificed after 6 hours.The tetramethylpyrazine(TMP)treated group showed that the WBC count was significantly higher than RDS group's at 15 min after fat tissue extract was given(P<0.05).In addition,the edemas of both capillary endothelial cells and alveolar epithelial cells were not as serious as that of RDS group.The numbers of accumulating leukocytes in the lungs were less than that of RDS group.The increase in production of leukotriene B_4(LTB_4)by polymor- phonuclear neutrophil(PMN),alveolar macrophage(AM),and the activity of platelet-activating factor(PAF) by PMN were partially inhibited(P<0.01).Although the pulmonary extravascular lung water volume/blood- free dry lung weight(PEWV/BFDL)of RDS group significantly elevated than that of control group,the treat- ment and control group's rate had no significant difference(P>0.05).However,the level of PaO_2,the PMN and AM count,and the albumin level of the BALF,of both treatment and RDS group,had no marked differ- ence.These results demonstrated that the early treatment of TMP could inhibit the decrease of the WBC count, and reduce the accumulation of leukocytes in the lungs.The primary beneficial mechanisms of TMP was related mainly to inhibiting leukocytic function and protecting the pulmonary capillary endothelial cells.
