摘要
应用骨髓移植动物模型及体外集落培养技术,观察了青蒿素琥珀酸钠(SA)对小鼠造血功能重建的影响。结果发现 SA(2~50mg/kg)腹腔注射,可明显促进骨髓移植小鼠 CFU-S 及 GM-CFU 形成,骨髓细胞及脾细胞数也有不同程度的提高;单独 SA 在体外不能促进正常小鼠 GM-CFU 的形成,但在重组小鼠 IL-3存在下,则明显促进 GM-CFU形成,提示 SA 可能通过与 IL-3的协同作用,加速骨髓移植小鼠的造血重建。此外,腹腔注射 SA 也能明显促进内源性 CFU-S 的形成,表明 SA 具有一定的放射保护作用。
By means of bone marrow transplantation animal model and in vitro colony culture technique,the effect of sodium artesunate(SA)on mice's hematopoietic reconstitution was assessed.The results demonstrated that SA (2-5 mg/kg)significantly accelerated the colony forming units of spleen(CFU-S)formation.increased the num- ber of GM-CFU.and nucleated hone marrow cell and splenocyte counts in bone marrow transplantation mice. SA alone in vitro had no effect on the GM-CFU of normal mice,however,in the presence of rm-IL-3.SA was able to increase the amount of GM-CFU significantly,suggesting that SA might accelerate the hematopoietic re- constitution by synergism with IL-3.In addition,SA intraperitoneal administered could markedly promote the production of endogenous CFU-S.indicating the radioprotective effects of SA.
