摘要
采用慢性苯中毒法建立大鼠再生障碍性贫血(再障)模型。观察再障大鼠红细胞中己糖激酶(HX)、醛缩酶(ALD)和丙酮酸激酶(PK)3个糖酵解关键酶活性的变化,以及益气生髓汤对其活性的影响。结果:再障大鼠红细胞的3种酶活性变化不显著。胃饲及腹腔注射给药均能明显改善再障大鼠的骨髓和外周血象。与未用药对照组比较,腹腔给药4周后,能明显维持再障组大鼠红细胞中 PK 活性(P<0.05)。提示:再障大鼠红细胞出现的能量代谢紊乱与其 HX、ALD 和 PK 活性无关,而益气生髓汤对再障大鼠确有一定疗效。
Aplastic anemia(AA)models were established in 34 rats by chronic benzene inhalation within 8 weeks. Forty-nine rats were divided into three groups:15 rats of normal control,18 AA rats and 16 AA rats treated with Yiqi Shengsui tang(YQSST).The RBC.WBC counts and hemoglobin(Hb)contents in their peripheral blood as well as the activities of hexokinase(HX).aldolase(ALD)and pyruvate kinase(PK)were determined before and after benzene inhalation and also following one course of YQSST therapy.YQSST was administered either intraperitoneally into 9 AA rats or orally to 7 AA rats with a dosage of 5 ml/kg body weight.Four weeks after benzene inhalation,RBC,WBC counts and Hb in the peripheral blood of AA rats showed a tendency of sig- nificant decline.The activities of HX,ALD and PK in the erythrocytes of AA rats revealed no significant change as compared with the normal control group.YQSST improved markedly the blood and bone marrow pictures of the AA rats,but failed to affect the activities of HX and ALD in the erythrocytes.However,its intraperitoneal injection exerted an obvious maintaining effect on the PK activity of erythrocytes in AA rats by the end of thera- py(P<0.05),when compared with AA rats in the same period,thus contributing somewhat to the recovery of energy level for the impaired erythrocyte glycolysis.The disorder of energy metabolism in AA rat erythrocytes was not related to the activities of their HX,ALD and PK.
