摘要
利用PCR技术从海南野生稻(OryzameyerianaBaill)核基因组中扩增出一个长度为393bp的DNA片段,并进行了克隆和全序列测定。由该基因片段推导的氨基酸序列与水稻(OryzasativaL。)胰蛋白酶抑制因子序列同源。将基因片段连接至表达载体pET21b并转化大肠杆菌8L21(DE3),经sDS-PAGE分析,检测到目的基因的诱导表达。
A DNA fragment 393 bp was amplofied by PCR technique from the genome of wild rice(Oryza meyeriana Baill)in Hainan Island and it was cloned and sequenced.The amino acid se-quence deduced from thegene fragment wa found homologous to that of trypsin inhibitor of therice Oryza sativa L. The gene fragment was ligated to the expression of vector pET2 1 b trans-formed into E. coli strain BL21(DF3).the induced expression the interested gene was detect-ed by SDS-PAGE analysis.
