摘要
观察在缺氧脑微血管内皮细胞培养液中,丙二醛(MDA)及乳酸脱氢酶(LDH)的含量变化与764-3对这一变化的效应。发现缺氧24小时脑微血管内皮细胞培养液中MDA含量有显著性升高(P<0.01),经764-3处理后缺氧的内皮细胞培养液中MDA含量处于正常水平。结果提示,764-3有抗氧化作用,但LDH在上述条件下无显著性变化。
Abstract Endothelial cells isolated from the calf cerebral homogenates were cultivated in saturated oxygen(95%O2+5%CO2)chamber.The MDA and LDH contents in the supernatants of the cell culture wer determined in normal and hypoxic(93%N+5%CO2+2%O2) condition for 24 hours.The MDA content of the hypoxic group was markedly increased to 11.72±2.95 pmol/L in comparing to the normal group of 6.78±2.25pmol/L.It indicated that lipid peroxidation had been taken place during hypoxia.Presence of 764-3(an effective component of Chinese herb Salvia miltiorrhiza in hypoxic culture could restore the production of MDA to the normal level of control group.There was no sign of more LDH release after the hypoxic insult.It suggested that hypoxia can induce lipid peroxidation in cerebral vessel endothelial cells and 764-3 is a potent antioxidant in this case.
