摘要
应用花粉管通道技术,将抗逆性强的长穗偃麦草总DNA导入普通小麦甘麦8号,后代中出现了广泛的变异,并筛选出两个高产、分蘖力强、抗条锈病的新品系。以这两个品系为材料,以供体和受体作为对照,研究了外源DNA导入后籽粒醇溶蛋白的变化,发现醇溶蛋白的增加、缺失和电泳迁移率的变化,新增加的组分与供体长穗偃麦草某些组分相对应。由此推测外源DNA导入受体后有可能某些DNA片段插入受体基因组从而导致受体基因表达改变或基因突变。
The DNA of Agropyron elongatum which is resistant to rust was introduced into commonwheat Ganmai 8 by the technique of anther tube passage,a great range of variations took place inthe progenies of the acceptor and two new lines with high yield,high tillering capacity and striperust resistance were obtained after selection.The gliadin of the two new lines were studied usingthat of the acceptor and the donor as control. It was found that some gliadins were added,somewere lost and some had changed in the character of electrophoretic mobility compared with theacceptor,in addition the gliadins added had their counterparts in Agropyron elongatum.It maybe concluded that some DNA fragments were inserted into the genome of the acceptor as the re-sult of introducing exogenous DNA,which would consequently lead to the mutation of some ac-ceptor genes or change the gene expression.
