rhGM-CSF cDNA直接注射小鼠骨骼肌的体内表达研究

摘要 本实验将含有人粒-巨噬细胞集落刺激因子(hGM-CSF)基因cDNA的重组真核表达质粒直接注射小鼠骨骼肌,观察了hGM-CSF基因在小鼠体内的分泌表达情况。ELISA结果显示注射重组质粒DNA后,第15天左右小鼠血液hGM-CSF的表达量最高,约有97 ng/ml,第20天次之,第25天和第10天的表达量相近,约有49 ng/ml。生物学活性检测结果表明,实验组小鼠血液有维持TF-1依赖株细胞的生长作用。表明重组质粒DNA直接注射骨骼肌不仅能表达hGM-CSF,而且表达产物有生物学恬性。 The plasmid pCG1 which was constructed by the eucaryotic expression plasmid pCDS, was directly transferred into the BAL B/c mice' s quadriceps. The results of ELISA showed that hGM-CSF was expressed and secreted in mice in vivo. The highest expression level of hGM-CSF was about 97ng/ml on the fifteenth day after DNA was injected. Meanwhile,the results of biological activity detection showed that C group mice's serum had the function of retaining the growth of TF-1 cells. This suggested that the hGM-CSF had certain biological function.
