摘要
用已建立的抗旋毛虫单克隆抗体杂交瘤细胞株(D_4,E_2)所分泌的单克隆抗体(McAb),经纯化后电泳分析,其重链分子量为55KD,轻链分子量为29KD,符合IgG抗体特点。两株单克隆抗体与旋毛虫阳性猪血清所针对的旋毛虫抗原决定簇相同,都不与猪的其它寄生虫发生交叉反应,与抗原的相对亲和力E_2>D_4,单克隆抗体亲和层析纯化抗原的分子量为49KD,经免疫斑点染色证明为糖蛋白。
The monoclonal antibodys (McAb) secreted by anti-trichinella spiralis hybrid-omas (D4,E2) which developed early in our lab were used in this study. After it was purified, McAb was used for electrophoresis and it was found that the molecular weight of heavy chain was 55KD, and the light chain was 29KD. It is accord with the trait of IgG. The two McAb and porcine serum positive to trichinella spiralis aim at the same determining cluster, and there are not crossing reaction between them and other parasites of porcine, relative affinity of E2 is stronger than D4. The molecular weight of McAb affinitive purified antigen is 49KD, and the immunitive spotted dyeing test of antigen showed that it is a glucoprotein.
