摘要 应用反转录一聚合酶链式反应(ReverseTranscription-PolymeraseChainReaction,RT-PCR)方法,亚克隆出大鼠肾脏血管紧张素Ⅱ-1型受体(AngiotensinⅡType-1Receptor,ATR1)cDNA,并以此为探针,通过Northernblotting分析和定量PCR方法,测定大鼠不同组织中ATR1mRNA的分布,证明ATR1mRNA广泛存在于不同组织中,其中以肾脏含量为最高,其次为心脏和小脑;自发性高血压大鼠(SpontaneouslyHypertensiveRat,SHR)肾、心组织中ATR1mRNA较正常对照大鼠(Wistar-KyotoRat,WKY)明显下隆。 ith reverse transcription-polymerase chain reaction(RT-PCR ),the complementary DNA of an-giotensin II type-1 receptor(ATRI ) had been subcloned from rat kidney. Expression and distribution of ATR1 gene in tissues in spontaneously hypertensive rat(SHR) and Wistar-Kyoto rat(WKY)had been de-tected by northern blotting and PCR,It has been found that ATR1 mRNA distributes widely in brain,heart,kidney,lung and liver in rat.The level of ATR1 mRNA is highest in kidney,followed in descending order by heart and cerebellum. In addition,this is the first report that ATR1 gene expression of heart and kidney of SHR is less than that of WKY.
