摘要
在以基因工程方法实现大肠杆菌表达外源重组人VEGF的基础上,应用制备性SDS—PAGE从包含体中分离纯化了这一因子。纯化蛋白在含有氧化型与还原型谷胱甘肽和肝素的缓冲液中复性,复性后的VEGF蛋白成为同源双体,经Miles方法检测,证明可提高血管通透性,具有生物活性。这一研究为制备具有生理与病理意义的研究奠定了基础。
n the basis of
expressing recombinant human VEGF in E. Coli we reported earlier,we
report the isola-tion and bioactivity recovery of this factor from
the procaryotic exprexsion products in this studyIsolated
re-combinant human VEGF is renatured in buffer containing oxidized
and reduced glutathione forms homodimer shown in non-reduced
analytical SDS-PAGE. Miles assay shows the renatured VEGF promotes
vessel perme-ability in dose-petency manner.
