摘要
将医用缝合线浸泡在质粒DNA溶液中,然后缝合在大鼠股四头肌上,可以观察到外源基因在肌肉组织中高效、稳定地表达,其表达量高于磷酸钙共沉淀和DNA沉淀块埋植法,较肌肉直接注射法表达量高10倍左右。

the efficiency of direct gene transfer in skeletal muscle.pN2-LacZ
plasmid DNA was dissolved in PBS containing 5%sucrose. Medical suture
was soaked in the plasmid solution for 24 hours before stitched on
the quadricep muscle,The same amount of DNA was transfered by calcium
phosphate coprecipitation implantation,DNA precipitation implantation
or direct injection into the quadricep muscle. LacZ gene expression
was detected by β-galactosidase assay and histochemical staining,and
the effi-ciency of foreign gene expression by stitching was much
higher than the other methods mentioned above.
