摘要
本研究连接合成了BQ123-多聚赖氨酸(BP)利用BQ123对血管平滑肌细胞(VSMC)膜表面内皮素A型受体(ETA)的高度选择性和多聚赖氨酸对DNA的强吸附作用,以BP携载含有β-半乳糖苷酶(β-LacZ)基因的表达质粒pSV-β-Galactosidase(pSV-β-LacZ)对培养的VSMC和内皮细胞(EC)转移基因。结果表明,BP具有向VSMC转基因性能,且依赖与靶细胞表面ETA的结合。
We constructed a soluble targeted DNA
delivery carrier,BQ123-Polylysine,by which plasmid DNA (pSV-β
-LacZ)was transfered into vascular smooth muscle cells(VSMC).The
expression levels of the re-porter gene(β-LacZ)in treated VSMC and
endothelial cells show that BQ123-Polylysine can efficiently
trans-fer DNA into VSMC,mediated by endothelin A subtype receptors of
VSMC surface.The results suggested that it is possible for
investigators to use this delivery carrier as a tool in research VSMC
gene regulation and cardiovascular gene therapy.
