香烟烟雾提取物对离体猪肺血管缺氧反应的影响

摘要 本文研究香烟烟雾提取物(CSE)是否可通过影响血管内皮细胞的功能从而改变肺血管对缺氧的反应性。发现CSE对肺动脉环的作用是先引起一短暂的收缩反应,接着发生舒张。收缩反应有内皮依赖性,但CSE舒张肺血管的反应无内皮依赖性。CSE引起的肺动脉环收缩反应不被阿托品、心得安、乙胺嗪、FPL55712等药物所抑制,但可被消炎痛、美蓝、棉酚所阻断。CSE可逆转HPV反应,这种逆转效应不被以上药物所阻断。结果提示:CSE的肺血管收缩效应有内皮依赖性。CSE引起肺血管内皮释放的是一种环氧合酶产物。CSE舒张肺动脉的效应是对平滑肌的直接作用。CSE可使离体缺氧性肺血管收缩反应转为舒张。 The present experiments were undertaken to determine whether the response to cigarette smoke extract in the isolated porcine pulmonary ring is endothelium-dependent.Changes in the tension of arterial rings were recorded isometrically.Removal of the endothelium was carride out by gentle rubbing and confirmed by Ach and morphologically.Treatment of unrubbed rings with cigarette smoke extract(CSE)caused a transient constriction followed by a vasodilation.The constrictive response was abolished by removal of endothelium,administration of indomethacin,methylene blue or gossypol.But the vasodilative effect of CSE remained in denuded rings. Hypoxic pulmonary vasoconstriction was reversed by CSE,this effect was not inhibited by the following agents:atropine,gossypol,methylene blue,indomethacin,propranolol diethylcarbamazine and FPL55712.The results snggested that,l)the contractive effect of CSE on pulmonary artery ring was endothelium-dependent,the endothelium derived constricting factor released in CSE-induced response might be a cyclooxygenase metabolite.2)the vasodilative effect of CSE was a direct one on pulmonary arterial smooth muscle. 3)CSE could reverse the hypoxic pulmongry vasoconstriction in porcine pulmonary artery in vitro.
