摘要
利用棉花枯萎病菌毒素作为选择剂,对经诱变的外植体而形成的愈伤细胞进行了筛选研究。不同的细胞群体需要不同的筛选压。诱变扩大了变异源,1.0~1.5K伦γ射线、0.15%~0.3%EMS处理4小时、1mmol/LNaN3处理2~3小时能有效地对外植体进行诱变。在一定的筛选程序下,获得了多个耐毒细胞系。耐毒细胞系及其再生株均表现出较好的耐毒性。
The toxin produced by Fusarium wilt of cotton was used as selecting factor toscreen tolerant cell lines from calli formed by mutated hypocotyle explants. Calli, suspensionsand embryoids needed different screening pressure. The variation range was widened by muta-tion. It is effective for 1.0 to l.5 krad γ─ray,0.15 to 0.3%EMS for 4 hours and 1 mmol/L NaN3 for 2 to 3 hours to mutate explants,respectively. Several cell lines tolerant to toxinwere screened out by an established screening procedure. The tolerance was kept in the regen-erated plants and calli subcultured on the medium free of toxin for one year.
