细菌感染后大鼠下丘脑促肾上腺皮质激素样和β-内啡肽样免疫反应阳性细胞的相应变化

The correlative changes between ACTH and beta-endorphin of the arcuate nucleus of the hypothalamus in the infected rats

摘要 采用Wistar大鼠,雌雄兼用,老龄鼠(20个月)9只,壮龄鼠(8个月)10只。经腹膜腔注入致病量福氏痢疾杆菌F1b(活菌492×104·g-1),24h后,处死取材,应用石蜡切片,ABC法染色,DAB显色。光镜观察下丘脑弓状核促肾上腺皮质激素样和β-内啡肽样免疫阳性细胞形态、分布和计数。采用图像分析仪测其灰度值。结果发现多数感染鼠第三脑室室周层出现许多β-内啡肽样免疫反应强阳性细胞。弓状核促肾上腺皮质激素样免疫阳性细胞数量老龄组显著低于壮龄组;β-内啡肽样免疫阳性细胞数量老、壮龄组相近。两类阳性细胞的灰度值老龄组均高于壮龄组。与正常大鼠相比较,感染老龄鼠弓状核促肾上腺皮质激素免疫阳性细胞灰度值显著上升,壮龄鼠则下降;感染组老、壮龄鼠弓状核β-内啡肽免疫阳性细胞灰度值均高于正常组之相应龄鼠。提示:促肾上腺皮质激素和β-内啡肽与抗感染密切相关。 In this study, the female and male wistar rats, 9 aged rats(twenty-month-old) and 10adult rats(eight-month-old) were used.Infected with shegella Flexner(Flb).injected in to cavum peritoneum, the rats were survived for 24 hours and were killed.The blocks containing hypothalamus were obtained, dehydrated and embedded in paraffin(50-52℃).The sections(7μm)were stained with ABC technology and coloured by DAB,The morphology, dis tribution and counting of adrenocorticotropin-immunoreactive(ACTH-ir) and beta-endorphin-immunoreactive(β-END-ir) neurons in the rat hypothalamus Were observed and measured under microscope. The light values of positive neurons Were measured with Image Analyser.We had found that there were many β-END-ir neurons of deep colour in the Ⅲ periventricular layer in the most of rats. The numbers of ACTH-ir neurons in the aged rats are lower than those in the adult rats, but the numbers of β-END-ir neurons, both aged and adult rats, are similar.The light values of ACTH-ir andβ-END-ir neurons in the aged rat arcuate nucleus are higher than those in the adult rat arcuate nucleus.Compared with the normal rats, the light value of ACTH-ir neurons in the infected aged rat arcuate nucleus is obviouly ascended. but is descended in the infected adult rat arcuate nucleus.The light values of β-END-ir neurons in the infected aged and adult rat arcuate nucleus are higher than those in the normal aged and adult rat arcuate nucleus.The results suggested that both ACTH andβ-END are correlated with infecting and immunizing.Complement words
