摘要
本文主要报导小豆初生叶、上胚轴、子叶、子叶节部位切段、主根根基部和主根根尖等外植体的愈伤组织诱导及其外植体的直接植物体再生实验结果。研究表明;初生叶以出芽后第二天的材料最好;而其它部位的外植体以出芽后第三天的愈伤组织分化诱导率较高。在发芽培养基中加入6—BA获得的供试外植体,其愈伤组织诱导所需时间短,而且诱导率均可达100%。,在6—BA浓度为0.1~0.4%的植物体直接再分化培养基中,上胚轴能够诱导出愈伤组织及芽、根、幼苗,并能顺利地驯化成植株;子叶也能诱导出愈伤组织、根和芽;其它外植体均可诱导出愈伤组织和根。幼胚在本实验的愈伤组织诱导培养基和再分化诱导培养基中既可诱导出愈伤组织,也可将原胚芽培养成苗。
This article mainly present the results of calli differentiation and direct plant regeneration
experiment on such explants from Adzuki bean as primary leaf,epicotyl, cotyledon,cotyledonary node, main
root base and the top point of main root,atc.The results show that the primary leaf two days after budding
is the best selection for tissure cultivation and the other explants can get a relativelg high induction
freqnency of calli differentiation on the third days of budding.The experimental ixplants in 6-BA budding
media take less time in its calli induction,and the induction freqnency can achieve 100 percent
commonly.In the direct plant redifferentiation media with a 6-BA concentration of 0.1
-0.4percent,epicotyl can develop calli,bud,root and seedling under induction,and it can grow into a plant
successfully;cotyledon can also develop calli,root,bud by induction;and other explants can also be induced
to grow calli and root under the same conditions. Yong embryo in both calli induction media and
redifferentiation media,can be induced to grow calli and original germ can also develop into seedling under
cultivation.
