本研究以组织化学方法观察离体断指组织中ALP、ATPase及SDH的变化,从功能角度观察组织活力而判断冷藏保护的有效程度与有效时限。结果显示血管壁细胞对氧最敏感。经过冷藏的肌肉与断指,合理的缺血时间分别为2与7d,可能争取的缺血时间分别为11与12d。
Tissue detached from living body will undergo degenerative process and finally lead to necrosis due to the deprivation of blood and oxygen supply. It has been proved already that cooling may lower metabolic process and protect the tissue from necrosis in a certain pe- riod of time. Storing the amputated digit (or extremity), composite tissue flap or transplants in a cool container to prolong the ischemic time limit is a proven effective measure. This ex- periment is designed to detect the effectiveness of cooling in delaying the degenerative pro- cess and to assess the reasonable ischemic time limit by biochemistry method. Ten amputated fingers were divided into tow groups and were kept in 15℃ incubator and 4℃ refrigerator separately. Specimens were taken from each group every 6 hours. The ALP, ATPase and SDH of the specimens were stain-determined and their staining density observed to assess the vitality of the tissue on a functional base. The results showed that the tissue of the vessel wall is more sensitive to ischemia (anoxia) than the skin and its appendages. The reasonable ischemic time limits for cool preserved muscle and amputated digit were 2 and 7 days respec- tively and they might be prolonges to 11 and 12 days respectively.
