用抗日本血吸虫虫卵单克隆抗体NP28-5B检测循环抗原的研究 被引量:3

摘要 本研究使用杂交瘤技术制备鼠抗日本血吸虫可溶性虫卵抗原(SEA)单克隆抗体NP28-5B,其同型鉴定为IgGl,针对的靶抗原分子量为140kD。将酶标记单抗NP28-5B进行Dot-ELISA直接法检测急性患者血清100份和慢性血吸虫病患者血清200份中循环抗原,阳性率分别为96%和98%,检测正常人血清200份,仅3例假阳性(1.5%)。用双盲法检测血吸虫病流行区江西矶山岛居民血清339份,其中粪检阳性血清114份,阳性率为89.47%,粪检阴性血清225份,阳性率为69.33%,研究证明NP28-5B具有高度诊断潜能。 In this study. a murine monoclonal antibody (MAb) NP28-5B against the soluble egg antigen (SEA) of Schistosoma japonicum was generated by hybridoma technology, Its isotype is IgGl. The characterization of antigenic specificity showed that 140kD molecules in SEA could be recognized by MAb NP28-5B with Western-blot. NP28-5B labelled with HRP was used in direct Dot-ELISA to detect circulating antigen in the sera of patients with acute and chronic schistosomiasis. The positive rate is 96% and 98% respectively, only three cases showing false positive reaction (1. 5%) in sera taken from 200 individuals living in nons-chis-tosomiasis area. In a field trial, a total of 339 serum samples from schistosomiasis epidemic area of Jishan Island in Jiangxi province was double-blindly examined by direct Dot-ELISA. Among them, the positive rate of 114 serum samples showing positive fecal egg examination was 89. 47% (102/114), and in 225 serum samples showing negative fecal egg examination, 69. 33% (156/225) also gave positive results, It is suggested that NP28-5B is of high potential for detecting the circulating antigen in the dignosis of schistosomiasis japonica.
