摘要
本文应用 ABC 免疫组织化学染色法结合葡萄糖氧化酶-DAB-硫酸镍铵(GDN)显色技术,观察了神经肽 Y(NPY)降钙素基因相关肽(CGRP)、P 物质(SP)和血管活性肠肽(VIP)免疫反应(Immunoreactire,IR)阳性神经纤维在猴脾脏的分布。结果发现,NPY-IR 神经纤维沿脾的动脉、静脉及其分支走行,在被膜、小梁、白髓、红髓和边缘区亦有分布。CGRP-IR 和SP-IR 神经纤维在脾脏的分布相同,主要沿脾动脉走行,中央动脉周围尤为丰富,白髓和红髓的淋巴组织中有少许分布。VIP-IR 神经纤维主要分布在中央动脉周围,白髓和边缘区有零星分布。猴脾脏肽能神经纤维末梢与免疫细胞接触密切,揭示它们对免疫细胞的发育和活性具有调节作用。本研究为神经免疫调节机制提供了形态学依据。
By the use of immunohistochemical ABC method combined with glucoseoxidase—DAB—nickel enhancement techique the distribution of neuropeptide Y(NPY),calcitonin gene—related peptide(CGRP),substance P(SP)and vasoac-tive intestinal peptide(VIP)immunoreactive(IR)fibers in the spleen of ma-caca mulatta has been determined.The results showed that the nerve fiberscontaining NPY immunoreactivity were maninly distributed along the branchesof arteries and veins.They were also observed in the capsule,in the lymphoidtissues of white pulp,red pulp and marginal zone.Fibers staining for CGRP andSP completely overlapped indicating coexistance.They were present in perivas-cular plexus suppliing arteries.Some CGRP—IR and SP—IR fibers travelledbetween the lymphoid tissue of white pulp and red puIp.VIP—IR nerve fibersoccurred in the central arterioles.Only very rarely they coursed in the lymphoidparenchyma.The positive fibers were closely contacted with immunocytes, which suggested that they might regulate the development and activity of im-munocytes.These pathways may play an important neuroimmunomodulatory rolein the spleen.
