二株抗人原发性肝癌AFP—McAb杂交瘤的建立

摘要 本文报道从GHC—1及GHC—3分泌的AFP,经过常规方法的改进,获得AFP—McAb。以5pg/kg AFP免疫后的BAL B/C小鼠脾细胞与SP2/0小鼠骨髓瘤细胞融合,采用ELISA—Ⅰ、DIBA、RPHA—Ⅰ和杂交瘤细胞免疫荧光法,筛建了二株可分泌抗人原发性肝癌的AFP—McAb杂交瘤,定名为GAH_1及GAH_2。 RPHA—Ⅰ测试表明,GAH_1及GAH_2分泌的McAb,可抑制原发性肝细胞癌病人的腹水和血清,但对脐带血不能完全抑制。 AFP-McAb was obtained from AFP secreted from GHC-1 and GHC-3 by the modified conventional methods. After the fusion between the splenic cells from BAL B/C mice immunized with 5μg/kg AFP and mouse myeloma cells SP 2/0, then applying ELISA-I,DIBA, RPHA-I and immunoflourescence staining, the hybridoma cell strains were screened. We succeeded in establishment of two hybridoma cell strains capable secreting anti-human primary hepatocellular cancer AFP-MeAb. These two cell strains were named GAH_1 and GAH_2. RPHA-I study showed that the MeAb secreted by GAH_1 and GAH_2 reacted inhibited with sample of human primary hepatocellular cancer, such as aseites and serum of the patients, but incomplete inhibited with cord blood.
