摘要
抑郁症患者有不同程度的睡眠异常。这是早已肯定的事实,模拟人类抑郁症的动物模型,见于文献的已有20多种,而这些动物模型是不是也表现类似抑郁症患者的睡眠异常,有关报道很少。本文用重复弱束缚应激(Repeated Mild Restrained Stress.RMRS)的大鼠作为抑郁症动物模型,进行了行为和睡眠特征的观察。其结果如下:1.RMRS 6天的大鼠(n=7)单个 REMS 平均持续时间比正常对照组(n=6)明显延长(P<0.01)。其总睡眠量、REMS 总量、慢波睡眠等与正常动物无明显差异。与正常动物相比,RMRS大鼠有类似抑郁症的行为改变,如激惹、摄食少、体重下降、开野实验活动明显减少(P<0.01)。2.在 RMRS 的时,给动物连续肌注抗抑郁剂氯丙咪嗪(Clomipramine.CLI;n=7)6天,可以抑制动物的部分行为异常和 REMS 持续时间的增加。结果提示:RMRS 模型在行为特征方面较好地模拟了人类抑郁症。
Polysomnographic recordings and behavioural changes in an animal model of depression prepared by re- peated mild restrained stress(RMRS)have been examined in the present study.The results were as follows: 1.The average duration of rapid eye movement sleep(REMS)episodes in RMRS rats(n=7)was significantly in- creased as compared with those of the normal animals of the control group(n=6).If clomipramine(CLI.an antidepres- sant agent)had been injected intramuscularly before each restraining,these increases of REMS duration could not be ob- served. 2.In open field test,the RMRS rats less active.The number of area they entered and the number of rearings was sig- nificantly decreased as compared with the data from the control group.But CLI could not reverse the sedative effect of RMRS. 3.Behavioural changes of the RMRS rats were accompanied by a reduction in food intake and growth,and thus RMRS rats were obviously lighter than the control animals.The above mentioned findings suggested that RMRS rats have behavioural changes analogous to human depression,but alterations of sleep patterns are not significant.
