冬虫夏草对体外培养肾小管细胞庆大霉素毒性的影响 被引量:2

The protective effect of Cordyceps Sinensis against gentamicin-induced renal tubular cell toxicity in vitro

摘要 本文在以往整体动物实验和离体器官灌注证实冬虫夏草对氨基糖甙肾毒性损伤具有良好防治作用的基础上,采用肾小管细胞培养技术制作体外庆大霉素损伤模型,观察冬虫夏草对该模型细胞损伤的影响,并与国外常用的抑制庆大霉素细胞转运的保护剂氯化钙作比较。结果发现,冬虫夏草可减少庆大霉素损伤后肾小管细胞NAG酶释放(400mg/L庆大霉素时,虫草组19.17±2.01mu/L VS对照组38.83±1.55mu/L,P<0.01;800mg/L庆大霉素时,虫草组25.17±2.78mu/L vs对照组55.5±7.49mu/L,P<0.01)和乳酸脱氢酶释放,保护细胞Na^+·K^+-ATP酶。冬虫夏草作用优于氯化钙。 In vitro aminoglycoside nephrotoxicity model was induced by adding gentamicin (at the concentration from 200 to 1600ug/ml)directly to cultured rabbit tubular cells to observe the influence of Cordyceps Sinensis (CS) on the severity of cell injury. The results showed that CS could decrease the excretion of NAGase and LDH by tubular cells to culture medium 24h after cell exposed to gentamicin(200-800μg/ml) and the use of CScould antagonize the inhibitive effect of gentamicin on cell Na^+-K^+-ATPase. In contrast to calcium chloride. the adding of CS on medium did not competed with gentamicin to bind and transport into cells as indicating by the change of gentamicin concentration in culture medium 24h later, which mig-ht suggest that the mechanism of CS on protecting against gentamicin toxicity was not correlated with cell gentamicin transport.
