人工虫草对大鼠庆大霉素急性肾毒性损害的影响

摘要 本文在以往研究证实天然虫草对氨基糖甙肾毒性损害具有良好防治作用的基础上,以人工培植虫草替代天然虫草,观察它对体外培养肾小管上皮细胞增殖,对庆大霉素所致大鼠急性肾毒性损害及庆大霉素血药浓度的影响。结果表明,人工虫草可减轻庆大霉索对肾的损害,使大鼠尿 NAG 酶排泄量减少,保钠功能相对保存,血尿素氮、肌酐上升幅度小,其菊粉清除率高于单用庆大霉素对照组.人工虫草与庆大霉素联合应用既不影响庆大霉素血药浓度,也不减少肾皮质的庆大霉素积聚量,提示其保护肾脏的机理可能不是通过抑制庆大霉素转运和拮抗庆大霉素的药理作用。而主要是通过促进对损害、坏死的肾小管细胞再生和修复。 Cordyceps Sinensis,a Chinese herb drug,had been used successfully in our previous study to protect against aminoglycoside nephrotoxicity.To test whether the fermented product of the fungus seperated from Cordyceps Sinensis owns the same effect as to take the place of Cordyceps Sinensis for clinical practice,10 female rats receiving IP injection of 160mg.kg^(-1)/day for 7 days were treated by CSf,while 9 rats received gentamicin alone as control.It was found that CSf could decrease the excreation of urinary NAGase and prevent exacerbated loss of sodium from kidney which induced by gentamicin toxicity,Azotemia was less prominent in CSf-treated rat than that of control.The mechanism of CSf to ameliorate gentamicin nephrotoxicity might be related not to gentamicin transport but to enhancing the proliferation of tubular cells.
