摘要
正常人外周血淋巴细胞与兔抗钙调素(CaM)或患者抗CaM自身抗体阳性血清共育后,制备40~70nm超薄切片,用胶体金标记SPA处理,JEM-1200EX透射电镜观察,见淋巴细胞结构完整,胞膜、胞浆、胞核内可见散在的金颗粒分布,与正常人或正常兔血清共育的淋巴细胞,同法处理未见金颗粒沉着。提示抗CaM可进人活的淋巴细胞。
Normal human peripheral blood lymphocytes which were incubated with the rabbit anticalmodulin (anti-CaM)or human anti-CaM positive sera were observed by transmission electron microscope after making the 40~70 nm ultra-thin sections of lymphocytes and using protein A labeled gold probe.On electron microscopy,we found that the treated lymphocyte structures remained normal and the gold particles dispersedly distributed in the cytoplasm membrane,cytoplasm and nucleus.No gold particle distribution was found in the lymphocytes which were incubated with normal human or rabbit anti- CaM negative sera,suggesting that anti-CaM antibody may penetrate into the human living lymphocytes.
