摘要
为了探讨ANF对ET生物学效应的拮抗作用,我们进行了下述实验。在ET致小鼠死亡实验中,观察到大剂量ANF可显著延长小鼠存活时间;在ET致大鼠血压升高实验中,观察到ANF有明显拮抗ET的升压作用;在大鼠离体胸主动脉环灌流实验中,观察到ANF对ET引起的血管收缩呈浓度依赖性的舒张作用;在培养的血管平滑肌细胞(VSMC)中,观察到ANF有显著抑制ET的促VSMC钙内流作用。实验结果表明,ANF对ET生物学效应具有部分拮抗作用,ANF作为治疗与ET有关疾病的辅助药物,可能具有潜在的临床意义。
In order to investigate inhibitory effect of atrial natriuretic factor(ANF)on biological action of endothelin(ET),the in vivo and in vitro experiments were done.It was found that in the experiment of ET-induced death in mice,simultaneous treatment with large dose of ANF significantly prolonged animal survival time;and the administration of ANF to rat obviously inhibited its blood pressure elevation produced by ET.And ANF dose-dependently relaxed isolated perfused rat aortic ring,which was precontracted by ET.In cultured vascular smooth muscle cells,ANF markedly lowered ET- stimulated Ca^(2+) influx.The results showed that ANF partly antagonized ET biological actions, suggesting that ANF,as an adjuvant to treat diseases related to ET,might be of prespective significance on clinical application.
