摘要
提要用高纯度重组人IFN-Y免疫家兔,制成抗IFN-Y多克隆抗体,建立了双抗体夹心ELISA法。本法最小检出量为1.56IU/ml,特异性好,不精密度批内CV4.6%~7.2%,批间CV6.9%~10.1%,回收率96.7%~105.3%。30名健康成人外周血细胞经PHA诱导后上清中IFN-Y活性为7.1士4.2IU/ml。
Human interferon-gamma was detected by a sandwich enzyme immunoassay using an antirecombinant interferon-gamma polyclonal antibody. The assay was specific and could detect levels of IFN-as low as 1. 56 IU/ml. The intraassay and interassay coefficients were 4. 6~7.2% and 6. 9~10. 1% respectively. The analytical recoveries were 99. 1~105. 3% . Thirty normal aludts were determined for IFN-in supernatants of PHA-activated blood cell and the results was 7. 1±4. 21 IU/ml.
