摘要
将抗兔出血症病毒(RHDV)的 AA8-1和 CG4-1两株单抗(McAb)分别致敏双醛化人红细胞,混合后建立单抗反向间接血凝(McAb-RPHA)方法。其敏感性较常规血凝试验(HA)高256倍,最小检出量为0.214 ng/ml。此法对感染兔的心、肝、脾、肺、骨髓、血液等检出率均为100%;脊髓、淋巴结、肌肉等组织的检出率分别为62.5%,58.3%和37.5%。HA 的检出率除肝为100%外,其余均低于 McAb—RPHA,不能检出脊髓、淋巴结和肌肉样品中的病毒抗原。
A reverse passive hemagglutination(RPHA)test for detection of rabbit hemorrhagic disease virus(RHDV)was prepared by mixture of aldchydized red blood cells sensitized with anti-RHDV monoclonal antibodies(McAb)AA8-1 and CD4-1 respectively.The minimal dose of RHDV protein detected by McAb-RPHA was 0.214ng/ml.It was shown that this method was 256 times higher in sensitivity than HA test.Virus tissue samples collected from different sources were examined by McAb-RPHA and HA tests.RHDV antigenwas detected by McAb-RPHA from all of heart,liver,spleen,lung,bone marrow and blood samples from rabbitsinoculated with infected DJRK cell culture or infected naturally.The positive rates of mesenteric lymphadens,popliteallymphadens and muscles were 62.5%,58.3% and 37.5% respectively.Only liver samples were detected by HA testwith positive rate of 100%.None of mesenterie lymphadens,spinal cord and muscles were positive by HA test.
