小鼠睾丸细胞诱变与胞内 cAMP、cGMP 和 Ca^(2+)含量变化的关系 被引量:1

摘要 实验设5,10,15,20,25,30 mg/kg 6个剂量组,腹腔注射环磷酰胺(Cy),进行小鼠睾丸细胞染色体畸变试验,同时测定睾丸细胞内的 cAMP、cGMP 和 Ca^(2+)含量变化。结果证实环磷酰胺(Cy)对睾丸细胞具诱变性,并存在剂量依赖关系。6个染毒组的睾丸细胞(精原、精母细胞)染色体畸变率均显著高于对照组(P<0.01,P<0.05)。睾丸细胞内 cAMP 水平下降,cGMP 水平上升,与对照组相比有极显著差异(p<0.01)。cAMP 水平与 Cy剂量之间呈负相关(r=-0.966 6),cGMP 水平与 Cy 剂量之间呈正相关(r=O.912 3);cAMP/cGMP 比值随 Cy剂量增加而下降,且极显著低于对照组(P<0.01);25,30 mg/kg 两组睾丸细胞内的 Ca^(2+)含量显著高于对照组(P<0.01),其它各组无显著差异。 Malec mice were injected intraperitoncally with cyclophosphamide(Cy)at doses of 5,10,15,20,25,30mg/kg.The mutagenicity was detected using tests cell chromosomal aberration analysis.In the meantime,the contents ofcAMP,cGMP and Ca^(2+) in mutagenic cells were determined.The results indicated that Cy was mutagenic to the mousetestis cells and there was a certain relationship between dosage and response.The frequencies of chromosomal aberrationin six-treated groups were markedly higher than those of the control(P<0.01,P<0.05).In mutagenic testis cells,thecontent of cAMP reduced,while the content of cGMP elevated.They were remarkably different from the control(P<0.01).There was negative correlation between dosages of Cy and the content of cAMP(r=-0.9666),but a positiverelationship existed between dosages and the content of cGMP(r=0.9123);The value of cAMP/cGMP wassignificantly decreased with the increase of dosage when compared to the control(P<0.01).The contents of Ca^(2+) in 25mg and 30 mg/kg groups were obviously higher than that of the control,other groups had no distinct changes.
