摘要
采用分子生物学斑点杂交分析技术,对四氧嘧啶所致糖尿病鼠的肌肉及脂肪组织葡萄糖转运蛋白GLUT_1、GLUT_4基因mRNA表达情况进行了研究,结果:糖尿病鼠脂肪组织GLUT_4 mRNA表达减少(P<0.05),且胰岛素治疗可恢复其表达(P<0.05)。提示细胞GLUT_4基因表达的减少进而引起相应蛋白的减少可能是糖尿病胰岛素抵抗的原因之一。
The study was proceeded on the mRNA expression of the GULT1 and GLUT4 gene in the alloxan diabetic rats by the technique of dot-blot hybridization analysis. The results showed that the levels of the GLUT4 mRNA expression were reduced in diabetic rats while increased in insulin treated diabetic rats. This suggests that the reduced GLUT4 mRNA expression which could affect the quantity of its posttranslational products may be one of the causes of the insulin resistance.
