摘要
我们用 ^(32)标记的癌基因探针,通过 RNA-DNA 斑点杂交技术发现亚硒酸钠能抑制人肝癌细胞株 BEL-7402的 c-myc 表达,同时增强 v-fos 同源序列的表达,硒还能有效地抑制由致癌剂 MNU 和 BP 诱发大鼠成纤维细胞(Rat-1)癌基因 c-myc 的过度表达。NBT实验表明硒具有较强的促使人早幼粒白血病细胞分化的作用。以上结果提示硒的抑增殖促分化作用可能与调节癌基因的表达(myc 和 fos)有关。
Using RNA-DNA hybridization with ^(32)P-labeled DNA probes,we have found thatselenium inhibits c-mye expression in hepatoma cells (BEL-7402),while it increases expressionof the sequence homologue to v-los gene in those cells.NBT reductive test detects that humanpromyeloeytic leukemia cells could be differentiated strongly by selenium.Selenium also results insuppre-ssion of c-myc expression induced by treatment of rat fibroblast cells with N-methylnitrourea or benzapyrene.These observations suggest that the effect of selenium onproliferation and differentiation of tumor cells may be correlated with alteration in expression ofmye and fos genes.
