高效液相色谱法同时测定止痢宁片中穿心莲内酯和脱水穿心莲内酯含量

Content Determination of Andrographolide and Dehydroandrographolide in Zhilining Tablets by HPLC

目的建立止痢宁片中穿心莲内酯和脱水穿心莲内酯的含量测定方法。方法色谱柱采用Hypersil C18柱(250 mm×4.6 mm,5μm),以甲醇-水(50∶50)为流动相,检测波长为225 nm和250 nm,流速为0.8 mL/min,进样量为10μL。结果穿心莲内酯与脱水穿心莲内酯的进样量分别在0.020 5～0.123 0μg和0.080 8～0.484 8μg范围内与相应的峰面积呈良好的线性关系,方法平均回收率分别为98.30%和98.62%,RSD分别为1.17%和1.02%(n=6)。结论所用方法简便、快速,两组分分离良好,其他成分无干扰,测定结果准确可靠,可作为该制剂质量控制的有效方法。

Objective To establish an HPLC method for the content determination of andrographolide and dehydroandrographolide in Zhilining Tablets.Methods The Hypersil C18 column（250 mm×4.6 mm,5 μm）was used.The mobile phase consisted of methanol-water（50 ∶50）.The UV detection wavelength was 225,250 nm.The flow rate was 0.8 mL/min.External standard method was used.Results The linear range was 0.020 5-0.123 0 μg for andrographolide and 0.080 8-0.484 8 μg for dehydroandrographolide.The average recoveries were 98.30%and 98.62%with RSD 1.17%and 1.02%（n=6）,respectively.Conclusion The proposed method appeared to be simple,rapid,sensitive and accurste.It is available to the quality control of zhilining tablets
