摘要
本实验用氢过氧化亚油酸作用于整装培养的人脐带静脉内皮细胞,采用高锰酸钾固定液固定。结果表明:高锰酸钾固定液提取了内皮细胞的线粒体、内质网;并显示氢过氧化亚油酸引起生物膜系统主要是线粒体和内质网损伤的整体结构图像。为阐明脂质过氧化引起的内皮细胞损伤及其在动脉粥样硬化发病机理中的作用提供了新资料。
In this experiment, we first used the float membrane whole-mount culturemethod to study the peroxidized injury to human endothelial cells. Theendothelial cells in culture were treated with linoleic acid hydroperoxide,and fixed by potassium permanganate solution. Under TEM, injurious effectof lipid peroxide on mitochondria and endoplasmic reticulum of endothelialcells was systematically observed. The results revealed that linoleic acid hydro-peroxide mainly injured the biomembrane system of endothelial cells, especi-ally mitochondria and endoplasmic reticulum. As the concentration of linoleicacid hydroperoxide increase, mitochondria were distorted, dilated and rounded,and crista were distorted and disappeared. Endoplasmic reticulum were dilated,broken and disappeared. The results provide new data for explanation toinjurious effect of peroxidation on endothelial cells in atherosclerosis.
