急性人工高眼压对大鼠视网膜节细胞琥珀酸脱氢酶活性的影响 被引量:3

摘要 本实验向大鼠眼前房加压灌注生理盐水并维持使视网膜电图b波消失的临界眼内压90分钟。高压眼与对照眼视网膜铺片同时进行琥珀酸脱氢酶(SDH)组织化学反应。光镜下可见高压眼视网膜的节细胞SDH反应较对照眼的浅淡,且高压眼视网膜的视神经纤维较少显色.用图像分析仪对样本进行组织水平与节细胞水平的灰度值测定,显示在两个水平上高压眼视网膜的SDH活性都明显下降.本文就高眼压下节细胞SDH活性下降的原因进行了讨论. Twelve Sprague-Dawley albino rats (24 eyes) were used in this study. Nor- mal saline was injected into the anterior chamber of the experimental eye until the b-wave of the electoretinogram was extinguished, then maintained the critical high intraocular pressure (HIOP) for 90 minutes. Just a puncture, without HIOP, was given to the anterior chamber of the other eye of the same rat as a control Both retinas of the experimental and control eyes were whole-mounted and treated with succinate dehydrogenase (SDH) histochemical technique. The reacted products of SDH showed in either retinal ganglion cells (RGCs) or optic nerve fibers of the experimental eyes were lighter than those of the control eyes under microscope. Furthermore, the grey scale values of the reacted products of SDH on the matched ratinas were measured at corresponding microscopic fields and RGCs with an image analysis system. The differences of the grey scale va- lues between the experimental and control retinas were high statistically signi- ficant (p<0.01). The results demonstrated that the activity of SDH on the retinas of experimental eyes was obviously decreased. It may be due to the is- chemia and anoxia resulted from experimental HIOP.
