摘要
本实验以大肠杆菌(Escherichia coli)HB101(RP_4)作为供体菌,以多能硫杆菌(Thiobacillus versutus)作为受体菌,通过接合的方法将RP_4质粒转移到了多能硫杆菌中.接合频率为1.9×10^(-5),RP_4质粒的三个抗性基因(Ap^R、Tc^R、Km^R)在多能硫杆菌中均得到了表达.再将RP_4质粒从多能硫杆菌反向转移到大肠杆菌HB101菌中,接合频率为3.3×10^(-1)~7.3×10^(-1),三个抗性基因也均得到了表达.实验证明,多能硫杆菌对于RP_4质粒既是一个受体菌,也是一个很好的供体菌.
RP_4 plasmid was transferred into Thiobacillus versutus recipient from Escherichia coli donor harbouring RP_4 plasmid. The frequency of conjugation was 1.9×10^(-5), and three resistant genes (Ap^R, Tc^R, Km^R)of RP_4 plasmid were all expressed in Thiobacillus versutus. RP_4 plasmid was then transferred reversibly into Escherichia coli HB101 from Thiobacillus versutus with the frequency of conjugation from 3.3×10^(-1) to 7.3×10^(-1), three resistant genes were also expressed. It has been showed that Thiobacillus versutus is a good donor as well as a recipient to RP_4 plasmid.
